**Kelly Services is currently seeking a Scientist** **for a Direct Hire role in San Diego.**

**Job Title** : **Scientist - Biochemistry and In Vitro Discovery**

Pay: $80-90k depending on experience

We are seeking a Scientist-level candidates to implement general biochemical and molecular biological methods for the optimization and production of novel lasso therapeutics, as well as corresponding biosynthetic enzymes. Candidates must have relevant experience expressing and purifying proteins from a variety of organisms. A successful candidate will be expected to play a broad role in advancing our discovery platform. Specific areas of responsibility for this position will include, but are not limited to, propagation and manipulation a variety of bacterial organisms, isolation and characterization of lasso peptide pathway enzymes, and protein engineering to optimize pathway enzymes. Candidates will continuously interact with a multi-disciplinary team and make important contributions to our groundbreaking science in an exciting start-up environment where everyone makes a difference.

**Key Responsibilities** :

Culture and manipulate a variety of bacterial strains, including wild-type and engineered strains, with initial focus on gram-positive bacteria.

Design and build genetic constructs on self-replicating and integrating plasmids for heterologous overexpression of lasso peptide biosynthetic enzymes and pathways from various hosts.

Optimize protein expression and purify lasso peptide biosynthetic enzymes.

Develop biochemical assays to characterize enzymes and high throughput screening assays to rapidly monitor improved enzyme variants for enhanced production of lasso peptides of interest.

Develop methods for in vitro reconstitution of biosynthetic pathways and cell-free biosynthesis of lasso peptides for rapid production of analogs.

Acquire, evaluate, interpret, and present experimental results internally, at partner meetings, and at scientific conferences.

Execute safe laboratory procedures and always wear personal protection equipment in the labs.

Follow and help enforce all protocols associated with maintaining safe operations in the labs and offices during the COVID-19 pandemic.

**Qualifications and Skills** :

Ph.D. (or equivalent experience) in biochemistry, enzymology, molecular biology, microbiology, synthetic biology, or related disciplines (non-Ph.D. scientists with extensive relevant experience also will be considered). Industrial experience or postdoctoral work in these disciplines is desirable.

Skilled in overexpressing proteins from a range of bacterial strains (e.g., E. coli, Pseudomonas, Burkholderia, Bacillus, Streptomyces, etc.). Experience working with and genetically manipulating gram-positive organisms is desirable.

Proficient in recombinant DNA techniques and tools for constructing plasmids and vectors containing biosynthetic pathway genetic components e.g., DNA design, cloning, isolation, and sequencing).

Skilled in implementing and optimizing methods for protein production, isolation, and purification.

Expertise in developing biochemical assays for characterizing enzymes and enzyme complexes.

Proficient in bioinformatic analysis, BLAST search, sequence alignment, genomic database search and related methods and software.

Experience designing, refactoring, expressing, characterizing, and optimizing natural product biosynthetic pathways is desirable.

Strong publication history in peer-reviewed journals.

Ability to thrive in a fast-paced, dynamic, multi-disciplinary, team-driven work environment.

Must be detail-oriented, self-motivated, and able to work independently.

Excellent organization, strong interpersonal skills, excellent oral and written communication skills. Able to manage, organize, and critically evaluate data.
