We are currently seeking a **Scientist (Protein Purification/Production)** to work onsite with the National Institutes of Health in **Bethesda, MD.**

TASKS/SERVICES. The contractor shall:




+ Provide support for molecular/structural biology projects, as well as immunogen testing and analysis for an effective HIV vaccine and vaccines for other pathogens.

+ Prepare protein probes for B cell sorting and antibody isolation projects.

+ Perform B cell sorting using multicolor flow cytometry and single B cell sequencing to identify antibodies of interest.

+ Provide expression of viral glycoproteins, antibodies, immunogens and virus-like particles using mammalian expression systems; provide expression of immunogens in alternative expression systems.

+ Provide DNA vector manipulation including expression vector design, DNA/protein sequence analysis, DNA recombination, mutagenesis and plasmid preparation.

+ Provide biochemical analysis of proteins.

+ Maintain and generate stable cell lines.

+ Maintain tissue culture facility (maintenance of cell lines, cell culture mediums and supplies etc.).





REQUIREMENTS. The contractor must have:




+ Ph. D. in Biochemistry, Molecular Biology, Immunology or related discipline. Three (3) years of specialized experience plus a Masters degree is equivalent to a Ph. D. Five (5) years of specialized experience plus a Bachelors degree is equivalent to a Ph. D.

+ Minimum of two (2) years of experience in a related field.




+ Extensive experience with protein purification techniques

+ Affinity, Size Exclusion, Ion-exchange, Hydrophobic interaction chromatography etc.

+ Hands-on experience with biophysical characterization of proteins

+ Including but not limited to: size exclusion chromatography, dynamic light scattering, biolayer interferometry, surface plasmon resonance, differential scanning calorimetry

+ Experience with biotinylation and fluorescent labeling of protein probes for B cell sorting projects.

+ Experience with B cell isolation, FACS sorting, single B cell sequencing.

+ Experience working with mammalian cell culture for production of glycoproteins and antibodies




+ Basic knowledge of computer science, systems analysis, and systems programming.

+ Computer skills to include MS Word, Excel, and PowerPoint.

+ Experience with C, C++.

+ Sound knowledge of bioinformatics approaches to analyze.




+ A highly motivated Ph.D. person, able to follow explicit directions for protein production, but not required to carry out innovation.

+ Demonstrates prior experience working in a research laboratory environment; demonstrated ability to work as part of a team.
